Epitalon vs FOXO4-DRI

Epitalon is bioregulators; FOXO4-DRI is mitochondrial & longevity. They are compared here because they are frequently considered against each other, not because they act alike.

On published evidence alone, Epitalon is the better documented of the two (preliminary human). That is a statement about the state of the literature, not a recommendation.

Epitalon

Epitalon is one of the short peptide bioregulators characterised by Vladimir Khavinson, a series of di- to tetrapeptides each associated with a specific tissue. The proposed mechanism is direct interaction with DNA promoter regions to modulate transcription of tissue-specific genes, restoring protein synthesis in ageing or damaged tissue. It is the most studied compound in the series, associated in the original work with restoration of melatonin rhythm and with telomerase activation in cell culture. Nearly all supporting work comes from a single research group and has not been independently replicated under contemporary trial standards, which is the central limitation of the entire class and should be stated on every page in it.

FOXO4-DRI

FOXO4-DRI is a retro-inverso peptide, built from D-amino acids in reversed sequence, a design that resists proteolysis while preserving the binding surface. It interferes with the interaction between FOXO4 and p53 in senescent cells, releasing p53 to the mitochondria and triggering apoptosis selectively in those cells while sparing healthy ones. The widely circulated result is a mouse study reporting restored fitness, fur density and renal function in aged animals. There is no human data whatsoever.

Epitalon vs FOXO4-DRI: common questions

What is the difference between Epitalon and FOXO4-DRI?

Epitalon is bioregulators and FOXO4-DRI is mitochondrial & longevity, so they act on different pathways. Epitalon: A short peptide bioregulator directed at the pineal gland, from the Khavinson series developed at the St. Petersburg Institute of Bioregulation and Gerontology. FOXO4-DRI: A senolytic peptide designed to selectively kill senescent cells by disrupting the FOXO4-p53 interaction.

Which has stronger evidence, Epitalon or FOXO4-DRI?

Epitalon has the stronger evidence base (preliminary human, about 2 human studies) compared with FOXO4-DRI (animal models only).

Are Epitalon and FOXO4-DRI FDA approved?

Neither is FDA approved for any indication.
