Pentapeptide-18

Pentapeptide-18 is a leucine-enkephalin analog (Tyr-D-Ala-Gly-Phe-Leu) with D-alanine at position 2 for stability. It mimics the enkephalins by binding enkephalin receptors on the nerve cell, which is proposed to reduce neuronal excitability and calcium influx and so decrease acetylcholine release, moderating muscle contraction. Because it acts on a neuronal receptor while Argireline interferes with SNARE complex assembly, the two are frequently formulated together for an additive effect.

SNAP-8

SNAP-8 extends the Argireline sequence by two amino acids, more closely matching the native N-terminal region of SNAP-25. The longer sequence is claimed to compete more effectively for a position in the SNARE complex and so reduce acetylcholine release further. Manufacturer studies report greater wrinkle-depth reduction than Argireline; independent comparative data is thin, and the larger molecule faces the same skin-penetration constraint.

Pentapeptide-18 vs SNAP-8: common questions

What is the difference between Pentapeptide-18 and SNAP-8?

Both are cosmetic & topical. Pentapeptide-18: An enkephalin-derived peptide that reduces muscle contraction through a different pathway than Argireline, and is usually combined with it. SNAP-8: An elongated Argireline analog with two additional residues, marketed as a more effective version of the same mechanism.

Which has stronger evidence, Pentapeptide-18 or SNAP-8?

They sit at the same evidence level: preliminary human. Pentapeptide-18 has about 3 human studies and SNAP-8 about 2.

Are Pentapeptide-18 and SNAP-8 FDA approved?

Neither is FDA approved for any indication.
